FIELD: biotechnology.
SUBSTANCE: invention relates to food biotechnology, namely to biochemistry of milk and dairy products, and can be used to determine the intensity of oxidative modification of infant formula milk proteins and adapted milk mixtures intended for baby food. Method for assessing the level of oxidative modification of infant formula milk proteins involves determining the content of carbonylated proteins. 0.2 ml of the prepared infant formula is added to two test tubes, 1 ml of 20% TCA acid and 1 ml of 2M HCl solution are added to the control test tube, 10 mM – FeSO4 and 0.3 mM H2O2 are added to the experimental test tube, then the experimental test tube is incubated for 15 minutes at temperature of 37 °C and 1 ml of 20% TCA acid and 1 ml of 2,4-DNPH, both tubes are incubated at temperature of 19-24 °C for 1 hour, after which they are centrifuged for 10 minutes at speed of 3,000 rpm, the supernatant is drained from the test tubes, the precipitate is washed three times with a mixture of ethanol-ethyl acetate (1:1) and dried, adding 5 ml of 8M urea and boiling until complete dissolution of the precipitate, measuring the optical density of the test sample relative to the control sample at wavelengths of 270 and 363 nm, determining the extinction index of aldehyde-dinitrophenylhydrazones CADNPHuv at λ= 270 nm and extinction index of ketone-dinitrophenylhydrazones CKDNPHuv at λ= 363 nm, the antioxidant reserve coefficient of the infant formula is calculated.
EFFECT: possibility of using the method for determining the level of oxidative modification of infant formula milk proteins, reduced labour intensity of the method.
