FIELD: biotechnology.
SUBSTANCE: invention relates to engineering enzymology, molecular biology and biotechnology. Disclosed is a method of producing modified hyperthermophilic type I L-asparaginase from Thermococcus sibiricus, having SEQ ID NO: 4, which contains mutations D54G and T56Q. Method involves changing the structure of the initial type I L-asparaginase from Thermococcus sibiricus with SEQ ID NO: 1 by replacing amino acid residues in 1 and 3 position of 4-member highly conserved DSTL motif, where in 1 position the D/G substitution is made, in the third position T/Q. Also disclosed is a modified type I hyperthermophilic L-asparaginase from Thermococcus sibiricus, obtained using said method and having SEQ ID NO: 4.
EFFECT: invention enables to obtain type I hyperthermophilic L-asparaginase from Thermococcus sibiricus with improved catalytic properties.
