FIELD: biotechnology.

SUBSTANCE: invention relates to biotechnology, specifically to a genetic construct for expression of the N-terminal domain of CTCF protein fused with dCas9 and GFP proteins. Said chimeric proteins can be used to modify the topology of genomic loci and form chromatin loops. Proposed genetic constructs have optimized nucleotide sequences coding parts of the native human architectural protein CTCF but minimizing the side effects of the C-terminal domain of this protein.

EFFECT: co-transfection of these genetic constructs together with gRNA into a eukaryotic system enables to influence regulatory contacts of any genome locus.
