FIELD: medicine.
SUBSTANCE: invention refers to histology, regenerative medicine and can be used for elastic fibre separation from aortic tissue. Aortic fragments are crushed, pre-treated and washed in distilled water and centrifuged at 3,400 rpm for 5 minutes three times, soaking in formalin and treatment with potassium hydroxide solution with complete immersion of material for 24 hours at temperature 18-20 °C, washing with distilled water five times at temperature 18-20 °C, homogenisation by means of multiple passage of contents with Pasteur pipette, centrifugation at 1,600 rpm for 10 minutes with subsequent suspension, conducting at least five cycles of homogenisation and centrifugation of material, transfer of obtained suspension into graduated Eppendorf test tube with capacity of 1.5 ml, freezing at -80 °C and lyophilic drying at 0.080 mbar and -40 °C, with subsequent transfer of lyophilisate on double-sided tape for further analysis. Pre-processed aortic sample is subjected to enzymatic hydrolysis with 1% solution of type II bacterial collagenase with enzymatic activity 230 units/mg in a shaker incubator for 2 hours at temperature 37 °C and 250 rpm. Aortic sample is soaked in 10% buffered formalin for 2 weeks; 40% potassium hydroxide is used; homogenisation and centrifugation cycles are performed to achieve supernatant pH 7.0.
EFFECT: invention provides reduced material and time expenditures in the process of elastic fibre recovery from aortic tissue.
