FIELD: chemistry.
SUBSTANCE: invention relates to organic chemistry and specifically to a method of producing N,N'-bis(3-aminopropyl)butane-1,4-diamine (spermine) – an important endogenous metabolite having various useful biological activity, by treating 1,4-dibromobutane. Method comprises treating 1,4-dibromobutane with 3,4-dihydropyrimidino[2,1-a]isoindol-6(2H)-one in molar ratio of 1,4-dibromobutane : 3,4-dihydropyrimidino[2,1-a]isoindol-6(2H)-one=1:2 at 125 °C for 4 hours. Method then includes recrystallisation of precipitate from glacial acetic acid, treatment with 20 % solution of hydrochloric acid at 120 °C for 10 hours, evaporation to dryness at pressure 10 mm Hg, boiling the solid residue with acetone, filtering the precipitate and drying at pressure of 10 mm Hg.
EFFECT: disclosed method is simple, effective and enables to obtain spermine in the form of a mixed salt in two steps with a shorter duration and overall output of 78 %.
