ANTIBIOTIC EREMOMYCIN AND METHOD OF ITS PREPARING Russian patent published in 1997 - IPC

FIELD: antibiotics. SUBSTANCE: antibiotic eremomycin has the following elemental composition, %: C - 54.86; H - 6.14; N - 9.02; Cl - 2.40; equivalent by chlorine = 1479; value $$$ = 10.03 (c = 1, water). Absorption maximum in UV-spectrum in water at $$$ = 282 nm/water, $$$ = 38.4; absorption maximum in alkaline solutions at $$$ = 30 nm/0.1 N NAOH; maximal absorption frequencies in IR-spectrum (KBr): $$$ = 3400-3200; bands: 2900, 1670, 1500 and 1200-1000 $$$. Antibiotic is amphoteric compound, easily soluble in acid and alkali aqueous solutions, insoluble in organic solvents, easily crystallizes from water saturated with ethyl acetate, from aqueous solutions of propanol (80%) and ethanol (50%), precipitates as base from the concentrated aqueous solutions in weak alkaline medium, decomposes at temperature above 250 C. Antibiotic gives positive reactions for free amino-group, carbohydrate and phenolic hydroxyl. Method of preparing antibiotic eremomycin involves growing culture-producer Nocardia orientalis (strain INA-238) on nutrient medium containing carbon sources, organic and inorganic sources of nitrogen and mineral salts. Obtained native solution is collected on resin Amberlite XAD-2 and the end product is eluted with mixture acetone/n-butanol/water taken at ratio 1:1:1 at pH = 3.0 $$$ 0.5. Then product is precipitated from an aqueous concentrate with acetone excess and purified by chromatography at pH = 6.5 $$$ 0.1 on carboxyl resin KB-2 in $$$-form with 0.5 N ammonia solution followed by precipitation with acetone and crystallization from aqueous ethanol. EFFECT: improved method of preparing.
