FIELD: biotechnology, genetic engineering, medicine. SUBSTANCE: on the basis of regulatory sequences of gene BLG (sheep beta-lactoglobulin) hybrid genes BLG-hIFNg2 and BLG-hIFNg3 (size is 10000 base pairs) providing expression of hIFNg (human gamma-interferon) in mammary gland of transgenic animals were constructed. The obtained genes consist of -SalI-SnaBI-fragment (size is 4.7 thousand base pairs) that is 5'-flanking sequence of gene BLG; -SnaBI-AsuII-fragment (size is 1.7 thousand base pairs) that is a part of genome gene hIFNg; -AsuII-Bam HI-fragment that is (in the case of gene BLG-hIFNg2) the sequence involving part of the third and all fourth exon, the third intron and part of 3'-flanking sequence of gene hIFNg; and the case of gene BLG-hIFNg3: part of the third and all fourth exon of gene hIFNg, part of the sixth exon, the sixth intron, the seventh exon containing polyadenylylation signal, part of 3'-flanking sequence, part of the first exon, the second and the third exons, the first, the second and part of the third intron of gene BLG; -Bam HI-SalI-fragment that is part of polylinker pUC-18 and has the unique recognition site by restriction endonuclease Sna BI.
