SUBSTANCE: invention is related to recombinant plasmid DNA pgC1HDR, including a human C1-esterase inhibitor (C1-INH) minigene, flanked by genomic sequences of the mouse Serping1 locus, while the expression of the minigene is controlled by the mouse Serping1 gene promoter, the polyadenylation signal of the SV40 virus and the transcription terminator from the human SKIV2L gene. A method for constructing minigenes has been proposed, which consists of obtaining a plasmid construct carrying all the coding exons of the gene, flanked by intronic sequences that ensure correct splicing of the minigene, a method for detecting full-length transcripts of the C1-ING minigene and the human SERPING1 and mouse Serping1 genes, and a method for assessing the levels of transcripts of the human SERPING1 gene and the human C1-ING minigene using quantitative PCR and oligonucleotide primers for their implementation.
