FIELD: microbiology, biotechnology.
SUBSTANCE: sensitive organism with predetermined body mass is inoculated on high air passage mucous and after death its dead body cools down at environmental temperature 15-25°C (preferably at 20°C) for 3-4 hours. Then dead body is dissected and heart blood is sampled. Pasteurellas isolated two-by-two (bipolarly) in form of frank capsules are detected in blood. Simultaneously pasteurellas are isolated from nutritive broth by centrifugation and dilution in physiological solution (pH 7.0) in form of morphologically identical capsulated coccobacillus. Coccobacillus in own capsule obtained as facultative anaerobe are stored up to application step in anabiosis state at 20°C (optimum) for 10 days (optimum) or in liquid broth at 5°C (optimum) without accession of atmospheric air in filled, sealed and horizontally arranged Pasteur pipettes.
EFFECT: method for production of pathogenic pasteurella strains which are morphologically and virulent identical to starting ones and storage thereof for followed application.
