FIELD: medicine.

SUBSTANCE: preparation is coloured according to van Gieson, microscope image of the preparation is magnified in 400 or more times and digitalised, preparation background is separated at luminosity levels of 240-255 and inverted, and on the basis of histogram average image luminosity level is adjusted to the level of 185. Then the image is transferred to Lab colour model, so that areas with level of 20-50 are separated at the "a" channel, and areas with level over 70 are substracted in the "L" channel (Lightness). Separation of background, areas with luminosity within 20-50 and over 70, as well as average luminosity level adjustment by histogram, are performed with 256 image luminosity grades. Comparative collagen fibre area in histological preparation is defined by the formula where: Scomp is the comparative collagen area at the tissue slice, Scoll is the collagen fibre area, Stis is the preparation tissue area.

EFFECT: improved accuracy of comparative collagen fibre area definition in a histological preparation.
