FIELD: medicine.

SUBSTANCE: to make histological preparations, histological material is fixed. Simultaneously with fixation, pieces of organs are dyed with the help of new compositions - mixtures of methanol solutions of the main thiazine dyes, eosin, buffer components. Fixed and dyed pieces without washing and dehydration are poured into paraffin. Paraffin cuts are deparaffined, differentiated in mixture of ethanol and xylol at volume ratio from 2:7 to 7:2, are clarified in xylol and enclosed. Composition No.1 for fixation and simultaneous dyeing of organs pieces includes methanol-dissolved azure 2, eosin-potassium and components of buffer mixture. In composition No.2 components of buffer mixture are dissolved in mixture of blue eosin-methylene of Leishman type and blue eosin-methylene by May-Grunwald at volume ratio from 5:1 to 3:1.

EFFECT: reduced number of operations and considerable savings of ethanol, increased information value of histological preparations - detection of tissue granulocytes in cuts, mastocytes, hemopoetic cells at various stages of maturation along with tissue elements of epithelial, conjunctive, muscular and neural tissues.
