METHOD OF USING STEM CELLS IN TISSUE REPLACEMENT THERAPY Russian patent published in 2011 - IPC C12N5/00

FIELD: medicine.

SUBSTANCE: pluripotential stem cells are genetically modified. It is followed with ablation of residual undifferentiated stem cells with an antiviral preparation. The genetic modification of the undifferentiated stem cells is performed with using a bicistronic cartridge containing a suicide gene regulated a control element specific for the pluripotential stem cells containing an antibiotic resistance gene also regulated by the control element. The modified cells are selected in the presence of a related antibiotic both after the modification, and immediately before the introduction to a recipient. Then, the selected cells are introduced in a recipient's body directly without preliminary differentiation in vitro, then a therapeutic course with the preparation activating the suicide gene in undifferentiated cells is conducted.

EFFECT: invention allows clinically effective making up for cell mass of an affected organ or tissue and shorter period of expectation for potential recipients due to the absence of a pre-transplantation stage of stem cell differentiation in vitro, negated risk of post-transplantation teratoma development in the recipients.
