FIELD: medicine.
SUBSTANCE: reaction mixture containing a fluorescent substance is analysed for thermal radiation of the substance in the preset points with fluorescent radiation. A convection polymerase chain reaction (PCR) is preceded with a number of heating/cooling cycles with continuous storage of the fluorescence intensity and temperature values. The derived fluorescence intensity values during the convection PCR are corrected by subtraction of a background fluorescence which is a current temperature sample in the preset point from a fluorescence data array evaluated before the convection PCR, and by division into a modulus difference of the limit temperature samples of two intensities preset in convection cells for enabling a convection process.
EFFECT: reliable real-time standardisation of the convection PCR values that enables both qualitative, and quantitative analyses of nucleic acids.
