"ORGAN-ON-CHIP" DEVICE Russian patent published in 2014 - IPC A61F2/00 C12M3/06 B01L3/00

Abstract RU 2517046 C2

FIELD: medicine.

SUBSTANCE: group of inventions relates to medicine and biology and can be applied for cultivation, research and testing of test compounds on tissues, organoids and stem cell niches in a format of miniaturised integrated circuit. For this purpose claimed are: an autonomous device, a method of its production and application for testing test compounds on cell populations, a power unit, as well as methods of cultivating cells and organoids, a method of testing chemical compounds on the said cells. An autonomous "organ-on-chip" device contains reservoirs for medium supply, sections of growth of organs with cavities for organs, including those with a cavity for stem cells; reservoirs for medium discharge. The cavity for organs is connected with a reservoir for medium supply and a reservoir for medium discharge by means of microfluidic channels and can be connected and/or contain sensors. Cultivation of cells and organoids is performed by means of the claimed autonomous device by loading cell suspension of tissue cut into cavities for organs, sealing the said cavities, as well as testing test compounds on the cells and organoids with their further microscopic estimation and determination of medium parameters by the said sensors.

EFFECT: connection of a "cavity for organs" with a reservoir for medium discharge, combined with the device, controlled by sensors, ensures long-term cultivation of cells, including in a live organism, and possibility of moving the said device without disconnection of liquid channels from the external reservoir for discharge, which ensures autonomous functioning of the device and extension of range of its application in the cell technologies.
