FIELD: biotechnology.

SUBSTANCE: method comprises carrying out PCR in two reaction mixtures with hybridisation-fluorescent recording the results using appropriate oligonucleotide primers and probes on the DNA target "Med24" and "pCKF" of the following composition: the dforward primer Med24 RealTime-S GCCAGTGTGTGTCTAAA, the reverse Med24 RealTime-As CAACATTCGTCGCAAAG, and the probe Med24 Zond FAM-ACATTGTGCTGGACTCACAGCCCC-BHQ1, the forward primer pCKF RealTime-S AACCGCCTAAGCACTTTAT, the reverse pCKF RealTime-As CGTCAGGAACTCAACGAA, and the probe pCKF Zond FAM-ATCAGAGAGCATTTGAGCGGTTG-BHQ1. The typical and atypical strains of medieval biovar are separated as follows: the absence of hybridisation-fluorescent signal on both the DNA-targets "Med24" and "pCKF" in typical strains, the presence on both the DNA-targets "Med24" and "pCKF" in atypical strains, the presence of a DNA-signal on "Med24" and the absence on "pCKF" in the strains not belonging to the medieval biovar strains.
