FIELD: chemistry.
SUBSTANCE: invention relates to a novel method of producing a hyaluronic acid derivative containing an aldehyde group in position (6) of the glycosamine polysaccharide fragment. The method of producing an oxidised hyaluronic acid derivative includes reacting hyaluronic acid with 4-R-TEMPO and an oxidant in a protonic solvent, where R is hydrogen or acetamide. The oxidant used is sodium hypochlorite or trichlorocyanuric acid (TCC), wherein the hyaluronic acid is in the form of a free acid or salt. The protonic solvent is water or aqueous solutions of salts of general formula MX, where M is an alkali metal, and X is selected from a group comprising HCO3 -, CO3 2-, PO4 3-, HPO4 2-, H2PO4 - and halogens. The reaction of hyaluronic acid with 4-R-TEMPO and the oxidant is carried out at temperature in the range of -10 to 20°C, preferably at 0°C, for 5 minutes to 24 hours, preferably for 10 hours. Cross-linked hyaluronan derivatives can be obtained when diamines or compounds with three or more amine groups are used. Cross-linked derivatives can also be obtained by reacting an aldehyde with hyaluronan, substituted with an aminoalkyl group HA-alkyl-HH2.
EFFECT: obtained derivative can be used to bind amines, diamines, amino acids, peptides and other compounds containing an amine group, for example via reductive amination under the effect of NaBH3CN in an aqueous or aqueous-organic solvent.
