FIELD: medicine.
SUBSTANCE: method involves introduced an intracellular pH sensor, recording fluorescence intensity at two wave length within maximum absorption, and calculating a signal. The intracellular pH sensor is SypHer2 built-in protein. Its expression in tumour cells is provided by imbedding a respective gene into tumour cell genome. A resultant signal is ratiometric. An intracellular pH sensor signal is recorded in vivo in a living tissue from the 4th to 28th days of the tumour growth every three days. The fluorescence is recorded in the visible range with light-induced excitation at wave length 430 nm and 500 nm. The signal is received at wave length 540 nm and exposure 5 seconds.
EFFECT: more accurate measurements and a possibility of monitoring over the period of the tumour growth if exposed to different factors, including by selective localisation of the pH-sensor in the tumour, and the absence of measurement dependence on the pH-sensor concentration.
