FIELD: biotechnologies.

SUBSTANCE: method of obtaining of the patient (donor)-specific fibroblast-like cells from induced pluripotent human stem cells is described. Clusters of induced pluripotent stem cells are mechanically dissociated down to monocellular culture in presence of dispase. The cell suspension is placed in the medium consisting of the medium for cultivation of undifferentiated ES cells (mTeSr) + medium for cultivation of embryoid bodies (EB) (ET medium) in the ratio 1:1. Then embryoid bodies are obtained by the hanging drop method or by placing of cells into 96-pit round-bottom pad. Then three-day EBs are transferred to 24-pit die covered with 0.1% gelatine, one body per pit in 1 ml of medium for EB. They are cultivated until appearance of the monolayer of fibroblast-like cells after 8-10 days with preliminary removal of residue of the embryoid body.

EFFECT: invention allows to obtain donor- and patient-specific fibroblast-like cells for their use in transplantology and as autologous feeding layer for cultivation of induced pluripotent human stem cells and their maintaining in undifferentiated state.
