FIELD: biotechnology.

SUBSTANCE: wet chamber, a standard tipless cantilever, a lymphocytes suspension is used. Suspension of lymphocytes is prepared by whole blood centrifugation without washing in phosphate buffer, cells viability is checked in vitro tests. A drop of 2% gelatin solution is applied to the center of the glass surface, and a drop of leukosuspension and a drop of erythrocyte suspension are placed on opposite edges. Then the tipless cantilever is immersed into a drop of 2% gelatin solution, then the scanning area is transferred to the edge of the glass surface where the leukosuspension drop is located, the site with separate lymphocytes is selected under the control of the microscope optical system, the tipless cantilever is drawn to the individual lymphocyte. After lymphocyte adherence to the tipless cantilever, leukocytes granular forms are scanned, then the scanning region of the sample is transferred to the edge with the erythrocyte suspension drop and scanned. The preparation of the biomechanical sensor and measurement of adhesion forces in the "cell-cell" system is carried out in the power spectroscopy mode during a single scanning.

EFFECT: use of the method allows to manufacture biomechanical sensors, which allow obtaining of high accuracy objective data on interaction forces in a cell-cell system.
