FIELD: medicine.

SUBSTANCE: group of inventions can be used to obtain an isolated cell population comprising non-graft-versus-host (GVHD) inducing cells against a third party having a central memory T-lymphocyte phenotype (Tcm). The method comprises: (a) treatment of non-sticking peripheral blood mononuclear cells (PBMCs) with an agent capable of depleting CD4+ and/or CD56+ cells to produce CD8+T cells; (b) contacting of the said CD8+ T cells with a third-party antigen or antigens in the presence of IL-21 for 12 hours to 5 days to enrich the antigen-reactive cells; and (c) culturing of the said cells obtained in step (b) in the presence of IL-21, IL-15 and IL-7 in an antigen-free medium for 5-20 days to allow proliferation of cells having the said T-lymphocyte phenotype central memory (Tcm), thereby obtaining an isolated population of cells. The group of inventions also relates to versions of the method for production of the above cells and to methods for treatment of a subject in need of cell or tissue transplantation as well as transplantation of immature hematopoietic cells. These cells against the third party have a central memory T-lymphocyte phenotype, including CD3+, CD8+, CD62L+, CD45RA-, CD45RO+ signature, wherein at least 50% of the isolated cell population is CD3+CD8+ cells.

EFFECT: application of this group of inventions allows to use a population of cells having a central memory T-lymphocyte phenotype and capable of homing to the lymph nodes after transplantation to induce tolerance.
