FIELD: biochemistry.
SUBSTANCE: invention relates to the field of biochemistry, gene engineering and biotechnology, in particular to a method for producing heterotetrameric recombinant streptavidin. Present method involves the creation of two plasmids, one of which encodes streptavidin of the mutant type and the other is wild-type streptavidin, transformation of the E. coli cells by said plasmids, induction of streptavidin synthesis, isolation of the periplasmic cell fraction and purification of heterotetrameric streptavidin on an affinity sorbent. Method is characterized in that in one embodiment, wild-type streptavidin is linked to an affinity tag, and mutant-type streptavidin is coupled to PelB leader peptide, in the second case, mutant-type streptavidin is linked to an affinity tag, and wild-type streptavidin is coupled to PelB-leader peptide.
EFFECT: present invention makes it possible to obtain heterotetrameric recombinant streptavidin with high efficacy.
