FIELD: medicine.
SUBSTANCE: invention relates to medicine, namely to regenerative medicine, and can be used to evaluate the functional properties of the tissue engineering design of the diaphragm in an experiment. To do this, a rat diaphragm is used, the resulting matrix is recellularized by applying a suspension of allogeneic multipotent mesenchymal stromal cells of bone marrow origin. After confirming the in vitro adhesion, viability, cell proliferation, and their ability to direct differentiation, the resulting design is orthotopically implanted in the rat at the site of the previously modeled defect of the diaphragm. After 21 days in vivo, functional tests are carried out: spirometry, electromyography, X-ray examination, computed tomography, registration of gas composition of blood and acid-base balance, determination of contractility of muscle tissue. Pathomorphological study of the explanted graft is also carried out. When identifying the indicators of the study, comparable to those in the functioning of the native tissue of the diaphragm, the functional compliance and full participation of the restored diaphragm in the act of breathing is established.
EFFECT: method provides comprehensive analysis of the quality of the tissue engineering design of the diaphragm.
