FIELD: biotechnology; medicine.
SUBSTANCE: group of inventions relates to biotechnology and medicine, namely, to a vector-free microfluidic platform for direct-to-cytosol intracellular delivery of compound or composition, to a eukaryotic cell. Provided are microfluidic systems for causing perturbations in a cell and methods for delivering a load into a cell. Microfluidic systems comprise a device, in particular a microfluidic channel comprising a cell-deforming constriction, and a component passing cells. Diameter of the constriction is 20-99% of the diameter and induce perturbations of the cell large enough for a payload to pass through. Method for delivering a payload into a cell, by using said system, is used to deliver DNA, RNA, siRNA or protein to primary fibroblasts and stem cells, antigen or RNA to primary immune cells, quantum dots, carbon nanotubes or drugs to the target cell.
EFFECT: inventions exclude modification of the desired payload, are suitable for delivery of sensitive loads, and also provide direct-to-cytosol delivery, avoiding the endosomal compartment, do not lead to a change in the differentiation state or treated cell activity.
