FIELD: biochemistry.

SUBSTANCE: invention relates to biochemistry. Antibody is described to increase the amount of antigen binding by the antibody, to improve the antibody retention in the plasma or to promote the capture of an antigen by the antibody in a cell comprising a Fab fragment and a human FcRn-binding domain, antigen binding activity of which differs in two different conditions of calcium concentration and is lower in conditions of a low concentration of calcium than under conditions of a high concentration of calcium, where the low concentration of calcium is the concentration of ionized calcium from 0.1 to 30 mcM, and the high concentration of calcium is the concentration of ionized calcium from 100 mcM to 10 mM, wherein said antibody comprises at least four amino acids selected from a group consisting of amino acids at positions 30, 31, 32, 50 and 92 according to the Kabat numbering in a light chain that have a metal chelating activity. Also, a pharmaceutical composition comprising the antibody is described.

EFFECT: invention can be used to accelerate the capture of the antigen by antigen-binding molecules in cells, increase the number of times of the antigen-binding by one antigen-binding molecule, accelerate the decrease the concentration of the antigen in the plasma, and increase the retention of the antigen-binding molecule in the plasma, as well as antigen-binding molecules.
