FIELD: biotechnology.
SUBSTANCE: following is presented: a method of producing an antibody which involves selecting an antibody that contains an antigen-binding domain and a human FcRn-binding domain, wherein the antibody has binding activity to human FcRn at pH 5.8 and 7.4 and lower antigen-binding activity at pH 5.8 than at pH 7.4, where the binding activity to human FcRn at pH 7.4 is higher than that of intact human IgG. Then, the host cell is cultivated, the said cell contains a vector into which a polynucleotide encoding the antibody selected in the previous step is introduced, and the antibody is isolated from the host cell.
EFFECT: invention makes it possible to obtain antibodies that have an activity to bind with human FcRn at plasma pH and lower antigen-binding activity at early endosome pH than at plasma pH; such antibodies can increase the number of antigens to which one antibody molecule can bind.
