FIELD: medicine.
SUBSTANCE: invention relates to medicine, namely, immunology, and can be used to produce a specific protein-containing antigenic preparation from clinically significant yeast fungi. To this end, yeast from yeast species Candida albicans, either Geotrichum candidum, either Rhodotorula mucilaginosa, either Cryptococcus neoformans, either Trichosporon cutaneum is cultivated in a liquid synthetic nutrient medium. Cells are then separated by centrifugation and filtration, and the cell-free culture fluid is lyophilized, standardized for protein concentration and is used in immunochemical reactions to determine the level of specific antibodies to these mycosis pathogens. Analyzes are carried out by precipitation, ELISA with blood serum of patients with candidiasis. Prior to standardization, the culture fluid is further freed from polysaccharides and glycoproteins by repeated freezing-thawing and molecular filtration is carried out through a molecular filter with a pore diameter of 100 kDa. Specificity of the antigen preparation obtained is estimated under one of the conditions that: fraction of antigens with a molecular mass below 100 kDa from yeast species Candida albicans, either Geotrichum candidum, either Rhodotorula mucilaginosa, either Cryptococcus neoformans, either Trichosporon cutaneum interact only with homologous sera in patients with candidiasis, but does not interact with the sera of patients with candidiasis of another kind of yeast fungi; the levels of specific antibodies to the antigenic preparation in patients are higher than in the control group of patients without candidiasis.
EFFECT: use of this invention makes it possible to obtain specific protein-containing preparations from several genera of clinically significant yeast fungi, which makes it possible to determine the level of specific serum and epithelial immunoglobulins in the differential diagnosis of mycoses and mycoallergoses.
