FIELD: food industry.
SUBSTANCE: invention relates to an assessment of the safety of food products, namely a method for quantitatively determining the content of okadaic acid (diarrhea toxin of mollusks) in seafood by HPLC-MS using an Agilent 1200 HPLC System liquid chromatograph and a Thermo Scientific Orbitrap Elite high-resolution mass spectrometer. To this end, the seafood is homogenized, an aqueous solution of methanol is added and then centrifuged and the supernatant containing the extract of okadaic acid is withdrawn. Extract composition was determined chromatographically on a C18 column in a gradient mode with eluents A (0.1 % solution of formic acid in water) and B (acetonitrile) and chromatogram registration. Detection of okadaic acid is carried out by ionization with electrospray in the positive mode of the mass spectrometer according to the intensity of the ion [MH-H2O]+, or [MH-2H2O]+, or [MH-3H2O]+. Calibration curve is used to quantitatively determine the content of okadaic acid in the samples. Lower detection interval is 5 to 20 mcg/kg with a measurement error of not more than 15 %.
EFFECT: invention provides quantitative determination of okadaic acid in seafood for examination and state supervision of food safety.
