FIELD: food industry.
SUBSTANCE: invention relates to the food industry. A method for quantitative determination of the residual content of steroid hormones is proposed: estrone, dihydrotestosterone, methyltestosterone and mestanolone in fish, which consists in homogenizing fish meat, adding distilled water to samples with their treatment in an ultrasonic bath for more complete extraction of hormones from homogenized substance, extraction of hormones from samples with 1% formic acid solution in acetonitrile, purification of aliquot silica gel with grafted octadecyl groups C18 and anion-exchange sorbent based on primary and secondary PSA amines, hormone derivatization with hydroxylamine solution, evaporation of samples on a rotary evaporator. Next, the samples are re-dissolved in acetonitrile, chromatographic separation using a chromatographic column 2.1×75 mm with a grain size of 3.5 microns filled with a reversed-phase sorbent C18, a water-methanol mixture with the addition of 0.1% formic acid is used as the gradient of the mobile phase. Then the hormone content in the samples is analyzed on a triple quadrupole mass detector with an electrospray ionization source in a positive mode.
EFFECT: method provides an increase in the sensitivity of the determination method.
