METHOD FOR PREPARING A NOOTROPIC COMPOSITION BASED ON POLYPEPTIDE COMPLEXES ISOLATED FROM NEURONS AND GLIAL CELLS OBTAINED BY DIRECTIONAL DIFFERENTIATION OF INDUCED HUMAN PLURIPOTENT STEM CELLS Russian patent published in 2019 - IPC A61K38/17 C12N5/07 A61K35/545

Abstract RU 2690498 C1

FIELD: biotechnology.

SUBSTANCE: invention relates to a method for producing a protein-peptide composition with nootropic properties and involves: collecting a skin biopsy material, obtaining a fibroblast culture, cryobreservation of the fibroblast culture, defrosting the fibroblast culture, reproducing the fibroblasts to produce induced pluripotent stem cells (IPSC), culturing IPSC, cryopreservation IPSC, defrosting IPSC, neuronal differentiation of IPSC, production of a protein-peptide complex. Producing a protein-peptide complex using such a method enables avoiding immunological conflicts, the development of which is possible when using peptide compositions of xenogenic origin derived from brain tissues of pigs, sheep and other animals. Besides, use of human cell cultures enables to obtain a protein-peptide composition containing proteins and peptides with molecular weight up to 100 kDa, which will include not only small peptides, but full-length protein neuronal growth and differentiation factors. Effectiveness of such factors is certainly higher than that of short peptides, and immunogenicity is absent.

EFFECT: use IPSC as a protein-peptide composition production source, it is possible to solve the problem of donor material deficiency and ethical issues occurring when using embryonic material and human embryonic stem cell cultures for similar purposes.
