FIELD: biotechnology.

SUBSTANCE: invention refers to biotechnology, namely to unrestricted freezing of peripheral blood precursor cells. Method involves preparation of cryopreservation solution, for which solution Dimethylsulfoxide ("Sigma-Aldrich", France) of high degree of purification (≥99.99 %) with PP 0.200–0.220, under conditions of hypothermia, it is added to a solution for infusion of sodium chloride 2.25 g and water for injection to 250 ml in a plastic container to obtain DMSO of 10 % concentration and is mixed with a concentrate apheresis precursor cells of peripheral blood in equal proportions, freezing from plus 17 ÷ plus 25 °C to minus 80 °C in mode of three-stage program in electric refrigerators with set control temperature of isothermal cold adaptation at each stage of freezing. At the first stage, the biological object is held for 10 minutes in an electric refrigerator with a set temperature plus 4±1 °C, at the second stage, the biological object is transferred into a electric freezer with a preset temperature of cold adaptation minus 28±1 °C and maintained in these conditions for 25 minutes, at the third stage, the biological object is transferred into a freezer-storage with a specified temperature of minus 80±2 °C for final freezing and prolonged cryopreservation of PCPB.

EFFECT: invention increases efficiency of freezing suspension of precursor cells of peripheral blood.
