FIELD: medicine.
SUBSTANCE: cryopreservation of blood HSC is carried out by stages. Initially, a cryoprotective 10% solution of the DMSO cryoprotectant is mixed with a concentrate of nucleated cells in an elastic package and the adaptation of the prepared cell suspension is carried out for 10 minutes at a temperature of 4±1°C, bio-object freezing from +4±1°C to -80÷-196°C is carried out in the fast two-stage program mode. At the first stage of freezing, cold adaptation is performed at a temperature of -26÷-30°C, with aging duration of 25 minutes, at the second stage, the bioobject is quickly transferred to a storage facility with a temperature of -80÷-196°C. Granulated heat transfer medium consisting of dry, disinfected Lab Armor thermogranules with a specified temperature of +4±1°C and -28±2°C, respectively is used as the refrigerant at the stages of cell suspension mixing with cryopreservant and cold adaptation of the hematopoietic stem cells.
EFFECT: method for hematopoietic stem cells cryopreservation improves the sterility and number of morphologically preservative nucleated cells.
