FIELD: biotechnology.
SUBSTANCE: invention relates to biotechnology and describes a new bacterial nuclease of CRISPR-Cas9 system from bacteria Defluviimonas sp. 20V17, as well as its use to form strictly specific double-strand breaks in a DNA molecule. Given nuclease has unusual properties and can be used as a tool for making changes in strictly defined places in a sequence of genomic DNA of unicellular or multicellular organisms.
EFFECT: invention makes it possible to increase universality of available CRISPR-Cas9 systems that will allow to use Cas9 nucases from different organisms for cutting of genome or plasmid DNA in more specific sites and under different conditions.
