MODULATING CELL GROWTH AND GLYCOSYLATION WHEN PRODUCING RECOMBINANT GLYCOPROTEINS Russian patent published in 2020 - IPC C12P21/00 C12N5/02

Abstract RU 2712562 C2

FIELD: biotechnology.

SUBSTANCE: group of inventions relates to biotechnology, particularly to methods of producing recombinant glycoprotein in fermentation culture in a CHO cell by controlling concentrations of iron, copper, zinc and manganese in the medium. In one embodiment, concentration of iron, copper, zinc and manganese is increased in serum-free culture medium at the beginning and/or during both growth phase and phase of fermentation product formation to increase biomass formation. In the second embodiment, concentration of zinc and manganese is increased, and concentration of iron and copper in serum-free culture medium at the beginning and/or during both growth phase and phase of product formation is optionally decreased to increase production of G0, G1 or G2 of recombinant glycoprotein glycoform. In the third embodiment (A), reducing the concentration of iron, copper, zinc and manganese or (B) increasing the concentration of iron and copper and reducing the concentration of zinc and manganese in the serum-free culture medium at the beginning and/or during the growth phase, and the fermentation process product formation phase to increase glycogen production of recombinant glycoprotein with high mannose content.

EFFECT: invention enables to affect production of mature N-glycosylated glycoproteins and/or immature N-glycosylated glycoproteins, mature non-fouled-isolated glycoproteins and/or immature non-fouled glycoproteins and/or to promote biomass formation.
