FIELD: chemistry.
SUBSTANCE: invention relates to biochemistry, specifically to production of a novel, modified form of thermostable protein GroEL of a microorganism T. thermophilus. Modified protein GroEL Thermus thermopiles has amino acid sequence of thermostable chaperon GroEL of T. thermopilus microorganism, in which residues of methionines Met164, Met209, Met286, Met314, Met489, Met541 are replaced with leucine residues. Also disclosed is a DNA fragment coding modified protein GroEL. Obtained modified protein GroEL is stable, thermostable and highly expressed in a host cell, is not subjected to proteolysis.
EFFECT: can be used in pharmacological industry as a carrier protein of desired peptides in fused protein structures by embedding DNA sequence coding target peptide, into a protein-carrier coding sequence for biosynthesis of recombinant peptides, synthesis of which is not cost-effective with existing technologies, in scientific research laboratories for producing different, having the required properties.
