FIELD: chemistry of polysaccharides.
SUBSTANCE: invention relates to the chemistry of polysaccharides. A bioactive composition based on a cross-linked hyaluronic acid salt containing quercetin is proposed. The average molecular weight of hyaluronic acid is 1800 kDa, quercetin in the composition is chemically bound to hyaluronic acid, at the same time being a bifunctional cross-linking agent for it. The composition is a dry powdery substance with the following ratio of components, wt%: quercetin - 0.5-1.5; hyaluronic acid - 98.5-99.5. Also proposed is a method for producing a bioactive composition based on a cross-linked hyaluronic acid salt containing quercetin. The method involves mixing powdered hyaluronic acid with an average molecular weight of 1800 kDa with quercetin powder for homogenizing the mixture in a mill at 10-35 °C for 10-15 minutes. Mechanochemical treatment is carried out by feeding the obtained homogeneous mixture of powders into a mechanochemical reactor of the twin-screw extruder type with sequential automatic movement into the feed zone and the second and third zones of mechanochemical treatment with the ratio of the lengths of the zones, respectively,%: 20:40:40. Mechanochemical treatment is carried out with simultaneous exposure to pressure within 700-800 MPa and shear deformation on cam mechanisms with a shear angle of 180 ° in total for 2-4 minutes in a nitrogen flow at a temperature in the feed zone of 5-10 °C, in the second mechanochemical zone - 100-110оС, in the third mechanochemical zone - 9-15 оС.
EFFECT: invention is aimed at obtaining a composition in which the macromolecular chains of HA are cross-linked with quercetin, as well as the development of an environmentally friendly method for preparing the composition, which makes it possible to obtain previously unknown cross-linked HA salts chemically modified with quercetin in a one-stage technological mode in the absence of a liquid medium without high energy, labor and water consumption, while receiving target products with a high yield.
