FIELD: biotechnology.
SUBSTANCE: invention relates to the field of biotechnology. A group of inventions is described, including an RNA replicon for protein expression, a trans-replication system for protein expression, including the above RNA replicon, DNA for use as a template for transcription in vivo or in vitro, a method for producing a protein of interest in a cell (variants), a host cell for producing a protein, a method for producing a protein of interest from a subject (variants). In one embodiment, the RNA replicon comprises a modified 5 'replication recognition sequence of an alpha-virus, wherein the 5' replication recognition sequence is modified by removing all translation initiation codons as compared to the native alpha-virus 5 'replication recognition sequence, wherein the alpha-virus is a Semliki Forest virus or a Sindbis virus.
EFFECT: invention expands the arsenal of tools for protein replication.
