RECOMBINANT GM PROTEIN CAPABLE OF BINDING Α2-MACROGLOBULIN AND APPLICATION THEREOF AS A LIGAND IN AFFINITY CHROMATOGRAPHY FOR EXTRACTING Α2-MACROGLOBULIN FROM HUMAN BLOOD SERUM Russian patent published in 2021 - IPC C07K14/315 C12N15/31 B01D15/38 C12N15/70 C12N1/21 C12R1/19

Abstract RU 2758604 C2

FIELD: biotechnology.

SUBSTANCE: invention relates to the field of biotechnology, specifically, to recombinant α2-macroglobulin (α2-M)-binding proteins, and can be used to isolate α2-macroglobulin (α2-M) from human blood plasma by means of affinity chromatography. A recombinant GM polypeptide is obtained from a group G streptococcus strain - G4223, capable of binding α2-M, IgG and HSA, and a pGM DNA encoding it, pQE 31-pGM recombinant plasmid DNA, and an E.coli M15-GM producer strain allowing expression of the recombinant GM polypeptide. Based on the recombinant GM polypeptide, an affinity sorbent is created wherein the GM polypeptide is attached to the matrix - cyanobromo-activated Sepharose 4B.

EFFECT: invention allows to improve efficiency of production of purified preparations of α2-M by eliminating labour-consuming process of preparing specific antibodies to alpha2-M, IgG and HSA, avoiding nonspecific "background" reactions often encountered in immunology, standardising the used G protein, isolating both alpha2-M and IgG and HSA from blood serum in one stage of purification.
