SUBSTANCE: invention relates to the field of biotechnology, in particular, to methods for editing the CXCR4 cell gene in order to prevent infection of human CD4 lymphoblasts with HIV-1 using the CRISPR/Cas editing system. The methods involve electroporation of CD4 lymphoblast cells with an electroporation mixture consisting of an effective amount of guide RNA, Cas9 endonuclease, and donor DNA, as well as knockin or directed insertion of a nucleotide sequence encoding a protective peptide of the gp41 group under the endogenous promoter of the CXCR4 gene.

EFFECT: invention is effective in treatment of HIV-1.
