METHOD FOR ADJUSTING THE CHEMILUMINESCENCE CURVE WITH A DECREASE IN THE CONCENTRATION OF THE CHEMILUMINESCENCE AMPLIFIER Russian patent published in 2022 - IPC G01N21/76

Abstract RU 2772505 C1

FIELD: biophysics.

SUBSTANCE: invention relates to the field of biophysics and can be used in the study of mechanisms and methods for regulating the processes of lipid peroxidation. The method consists in recording a series of optical density spectra of the reaction mixture using a spectrophotometer in the range of 300-600 nm, followed by determining the concentrations of substances at various points in the reaction time. The registration of a series of optical density spectra of the reaction mixture is carried out with the following parameters: the time of registration of the spectrum is 1 minute, the registration speed is 5 nm/s, the increment (step) is 0.5 or 1 nm. Registration of the first optical density spectrum should begin no more than 15 seconds after the start of the reaction, the time intervals between measurements are: 10 seconds from the first to the seventh measurement, 60 seconds from the 7th to the 30th measurement, 120 seconds from the 30th measurement. Based on a series of optical density spectra, the concentrations of the chemiluminescence amplifier are calculated at various points in the reaction time and the pseudo-first-order reaction rate constants are determined for the destruction of the chemiluminescence amplifier (activator). Correction coefficients for a decrease in the concentration of the chemiluminescence amplifier (activator) during the reaction time are equal to an exponent raised to the power of the product of the reaction rate constant of the pseudo-first order of destruction of the chemiluminescence amplifier by the time elapsed from the beginning of the reaction, if the destruction rate constant of the chemiluminescence amplifier for a given time is different from that in the first seconds of the reaction, then the described coefficient it is also multiplied by a technical coefficient (A) equal to the quotient of the unit and the value of the concentration fraction of the chemiluminescence amplifier, remaining in the system at a time equal to the first value of the time range for which the constant of the reaction rate of the destruction of the amplifier is relevant. The formula for adjusting the chemiluminescence intensity values has the form:

,

where k is the pseudo-first order reaction rate constant for the destruction of the chemiluminescence amplifier; t1 and t2 are the time from the beginning of the reaction corresponding to the initial and final boundary of the time range in which the constant k is adequate; e is the base of the natural logarithm; t is the time from the beginning of the reaction; Jc is the corrected value of the chemiluminescence intensity at time t; Je. its value, measured by a chemiluminometer, at time t; A is the technical coefficient, and t1 = 0 A = 1, at t1>0 A>1.

EFFECT: increase in the accuracy of the result and the reliability and adequacy of the data obtained.
