FIELD: biotechnology.
SUBSTANCE: invention relates to biotechnology, namely to recombinant plasmid providing biosynthesis and secretion of prochymosin. Recombinant plasmid pSF002 is proposed, providing biosynthesis and secretion of prochymosin with SEQ ID NO: 1, having a size of 6106 p.o. and consisting of following elements: a) AOX1 promoter – 5'-end domain of alcohol oxidase promoter; b) Alpha factor signal peptide – a section encoding N-end signal peptide ppMFα1D with SEQ ID NO: 12; c) a sequence of bovine prochymosin gene, optimized for expression in Pichia pastoris; d) tTEF1 transcription terminator; e) pUC origin – bacterial point of replication start pUC; f) Amp(R) – gene Amp providing ampicillin resistance; g) Zeo(R) – gene BleoR of resistance to selective antibiotic zeocin.
EFFECT: invention provides for effective synthesis of target protein.
