FIELD: physics.

SUBSTANCE: invention relates to structured lighting (SIM) microscopy. Such result is achieved by the fact that the optical pattern is projected onto the biological sample and the first image of the optical pattern applied on the biological sample is captured; phase-shifted projected optical pattern relative to the biological sample and capturing at least a second image of the phase-shifted optical pattern superimposed on the biological sample; and reconstructing a high-resolution image representing a biological sample, based on the first captured image and the at least second captured image, wherein the biological sample is contained in a structured flow cell comprising a plurality of nanosized holes having an elongated shape in the xy plane orthogonal to the projection direction.

EFFECT: technical result consists in reducing the number of images and dimensions required to allow fluorescent samples using SIM using structurally specially flow cells, and optimization of movement of light beam relative to fluorescent samples to achieve implementation of SIM, which can be used in methods of linear scanning.
