FIELD: physics.

SUBSTANCE: invention relates to techniques for reducing the number of angles required during formation of images in structured illumination of biological samples, by using flow cells with a formed pattern. Disclosed method of forming biological sample images comprises the following steps: directing light through a fixed optical diffraction grating, projecting an optical diffraction grating pattern generated by light directed through the fixed optical grating into a biological sample; performing a biological sample linear scanning; moving the biological sample relative to the pattern of the optical diffraction grating or moving the light directed through the fixed optical diffraction grating, and reconstructing the high-resolution image typical of the biological sample. At least part of the biological sample is placed in a plurality of nanosized holes, and said plurality of nanosize holes has orientation forming a square pattern.

EFFECT: technical result is reduction of number of images and sizes required for resolution (discrimination) of fluorescent samples using SIM-technology using application of special pattern of flow cells, as well as for efficient use of light beam movement relative to fluorescent samples in order to practically implement SIM technology, which can be used together with linear scanning technologies.
