SUBSTANCE: invention relates to the field of biotechnology. White-grain or red-grain or violet-grain soft wheat varieties are crossed with blue-grain varieties/lines carrying the ThMyc4E long wheatgrass gene, which controls the blue color of the grain aleurone layer, then DNA is isolated from the green leaves of the obtained second-generation F2 hybrids and analyzed using a set of PCR markers, consisting of the intragenic PCR marker TaMuc4E and microsatellite markers Xgwm0251/Xgwm0375 and Xgwm0624 mapped on chromosomes 4B and 4D of common wheat, respectively.

EFFECT: use of a set of PCR markers makes it possible to identify the presence of the ThMyc4E gene, as well as chromosomes 4B and 4D, in the genome of the obtained hybrids. Based on the presence of PCR fragments of each of the markers, a conclusion is made about the homozygous or heterozygous state of the dominant allele of the ThMyc4E gene in hybrids, as well as about the complete replacement of chromosomes 4B or 4D.
