FIELD: biotechnology.
SUBSTANCE: invention relates to the field of biotechnology, in particular to the production of 2'-fucosyllactose, using a genetically modified prokaryotic host cell. The host cell is genetically modified in such a way that at least (1) activity of enzyme converting fructose-6-phosphate is reduced or suppressed and/or by increase in activity of fructose-1,6-biphosphate-phosphatase; (2) at least one gene encoding enzyme necessary for synthesis of GDP-fucose de novo is super-expressed in the host cell; (3) an exogenous gene encoding alpha-1,2-fucosyltransferase is expressed in the host cell; and (4) a gene encoding fructose-1,6-biphosphate-aldolase is super-expressed. The specified host cell is cultivated in a cultural medium containing glycerin, and supply of lactose is provided. Fucosylated oligosaccharide is obtained, which can be extracted from the medium.
EFFECT: invention allows for biosynthesis of 2'-fucosyllactose in a concentration exceeding 100 g/l.
