FIELD: molecular biology; genetics; personalized medicine.
SUBSTANCE: method for determining genetic markers of the polygenic risk of developing dementias of the Alzheimer's type is presented. The method includes providing a biological microchip with immobilized oligonucleotide probes, multiplex amplification of fragments of the above genes, hybridization of amplified fluorescently labeled products on the biochip, registration and interpretation of hybridization results with determination of genotypes associated with the risk of Alzheimer's dementia. The method provides identification of 23 genetic markers including rs6656401 in CR1 gene, rs6733839 in BIN1 gene, rs35349669 in INPP5D gene, rs190982 in MEF2C gene, rs9271192 in HLA-DRB5/1 gene, rs10948363 in CD2AP gene, rs2718058 in NME8 gene, rs1476679 in ZCWPW1 gene, rs11771145 in ЕРНА1 gene, rs28834970 in PTK2B gene, rs9331896 in CLU gene, rs10838725 in CELF1 gene, rs7274581 in CASS4 gene, rs983392 in MS4A6A gene, rs10792832 in PICALM gene, rs11218343 in SORL1 gene, rs17125944 in FERMT2 gene, rs10498633 in PIN-SLC24A gene, rs8093731 in DSG2 gene, rs4147929 in ABCA7 gene, rs3865444 in CD33 gene, rs429358 and rs7412 in APOE gene.
EFFECT: method allows to quickly and accurately simultaneously identify genomic markers of the polygenic risk of developing dementia of the Alzheimer's type.
