FIELD: biotechnology; medicine.
SUBSTANCE: test system for the detection of nucleic acids of pathogens of CNS infections containing pairs of highly specific and sensitive binary probes represented by DNAzymes is described. Upon detection of the target nucleic acid of the causative agent of an infectious disease, specific binding to its sites occurs and a guanine quadruplex is formed, which has the enzymatic activity of peroxidase. This allows the pathogen to be detected by means of a colorimetric reaction. The probes are represented by oligonucleotides with the sequences SEQ 16 — SEQ 45 containing sites that recognize at least one of the nucleic acid sequences of pathogens in the biosample, and pathogens include sequences corresponding to those presented in SEQ ID 1 — SEQ ID 15. The test system allows to identify the following pathogens: N. meningitidis, S. pneumoniae, H. influenzae, E. coli, L. monocytogenes, S. agalactiae, Candida albicans, Mycobacteria tuberculosis, S. aureus, Herpes simplex virus 1, Herpes simplex virus 2, Varicella zoster virus, Cytomegalovirus, Epstein Barr virus, Herpes virus 6, Enterovirus. The invention is directed to the detection of a product accumulated by cyclic or isothermal PCR. The invention is adapted to work in a piston cartridge, microfluidic cartridge, biomanipulator with numerical control.
EFFECT: claimed test system is easy to operate, does not require any special laboratory conditions and professional skills, and provides a fast and reliable result.
