FIELD: chemical; pharmaceutical industry.
SUBSTANCE: invention relates to a method of quantitative determination of the amount of flavonoids in the leaves of the three-leaf watch. The method is as follows: the leaves of the three-leafed watch are grinded to a particle size of 1 mm, then a water-alcohol extract is obtained by a single extraction of 1 g of an accurate weight of the crushed leaves of the three-leaf watch with 70% ethyl alcohol for 45 minutes, at a ratio of raw materials:extractant 1:25, with subsequent sample preparation and determination of optical density by differential spectrophotometry using a standard sample of rutin, and in its absence — using a theoretical specific absorption index, quantitative determination of the amount of flavonoids in the leaves of a trifoliate watch using a color reaction with aluminum chloride solution of 5%, at a wavelength of 409 nm equivalent to rutin and the content of the total flavonoids equivalent to rutin and absolutely dry raw materials are calculated by the following formula:
where x is the content of the sum of flavonoids equivalent to rutin, %; D is the optical density of the test solution; Do is the optical density of a solution of a standard sample of rutin; m is the mass of raw materials, g; mo is the mass of the standard sample of rutin, g; W is the weight loss upon drying, %; in the absence of a standard sample of rutin, it is advisable to use the theoretical value of its specific absorption rate, equal to 248:
where x is the content of the sum of flavonoids equivalent to rutin, %; D is the optical density of the test solution; m is the mass of raw materials, g; 248 is specific absorption index of the standard sample of rutin at 409 nm; W is the weight loss upon drying, %.
EFFECT: above method allows to quantify the amount of flavonoids equivalent to rutin in water-alcohol extracts from the leaves of the trifoliate watch using differential spectrophotometry.
