SUBSTANCE: following is proposed: a method of modeling Mycobacterium tuberculosis biofilms in vitro, which consists in obtaining Mycobacterium tuberculosis biofilms in the interval between 7–30 days of static cultivation in the Middlebrook 7H9 liquid nutrient medium with an enriching additive Middlebrook ADC Enrichment, while pre-cultivation treatment of the inoculated M. tuberculosis suspension is carried out using incubation of the suspension in the presence of 25 mM/l methyl-β -cyclodextrin, or poloxamer-407 at a concentration of 2.0% w/v, or Tween-80 at a concentration of 0.1 w/v%, or tyloxapol at a concentration of 0.05% w/v, or n-octyl-β- D-glucopyranoside at a concentration of 0.5% w/v, or n-dodecyl-D-β -maltoside at a concentration of 1.0% w/v at a given temperature for 24 hours with constant stirring at 150–200 rpm (options).
EFFECT: inventions provide an expansion of the arsenal of methods of modeling biofilms of Mycobacterium tuberculosis for studying the physical, molecular genetic, biochemical, morphological and other characteristics of M. tuberculosis in the composition of biofilms and for testing their drug sensitivity to anti-tuberculosis drugs.
