FIELD: molecular biology.
SUBSTANCE: following method was proposed for isolating viral RNA from cerebrospinal fluid (CSF), which includes concentrating a CSF sample in a centrifugal vacuum concentrator with an acceleration of 150–300 g and a negative pressure of 2 kPa until the final volume of the sample is reduced by 4 or 5 times; adding in a ratio of 1:10 a lysis solution containing 4 M guanidine thiocyanate, 25 mM sodium citrate, 0.5% sodium lauryl sarcosinate, 0.1 M 2-mercoptoethanol, incubating the mixture until the components are completely dissolved; then 1-3 M sodium acetate, phenol and a mixture of chloroform and isoamyl alcohol in a ratio of 49:1 are sequentially added in a ratio of 1:10:2 with stirring; cool and centrifuge with an acceleration of 10–13 thousand g; the upper transparent aqueous phase is reprecipitated by adding isopropanol and incubating at minus 20°C; the resulting RNA is further purified by affinity sorption using silica.
EFFECT: invention provides an increase in the concentration and degree of purification of RNA when isolating it from biological samples of CSF with a low initial concentration of the pathogen.
