SUBSTANCE: method of producing the yeast Komagataella kurtzmanii with an improved ability to assimilate sorbitol, including transformation of K. kurtzmanii yeast cells with recombinant plasmid DNA pPH727-Sdh115, containing: a DNA fragment encoding the SOR1 gene (PAS_chr1-1_0490) of the yeast K. phaffii GS115, including the sequence of the structural gene sorbitol dehydrogenase from the yeast K. phaffii with a size of 1047 bp. and native 3'-UTR (3' -untranslated region) 211 bp long, defined as the promoter of the SOR1 gene, the terminator region of the alcohol oxidase gene AOX1 of the yeast K. kurtzmanii, the HIS4 gene of the yeast K. kurtzmanii, fragments of the distal part of the promoter region of the alcohol oxidase gene AOX1 of the yeast K. kurtzmanii, a fragment of plasmid DNA pUC18, ApR resistance gene, which ensures the selection of plasmid-containing E. coli cells on ampicillin-containing media; or transformation of K. kurtzmanii yeast cells with recombinant plasmid DNA pPH727-Sdh727 containing instead of the SOR1 gene (PAS_chr1-1_0490) of K. phaffii yeast (SOR1GS115) the SOR1 gene of K. kurtzmanii yeast (SOR1Y-727).
EFFECT: invention provides an expansion of the range of producers of recombinant proteins based on K. kurtzmanii, the fermentation of which uses sorbitol.
